The former Liverpool man was operated on last week, undergoing surgery to remove a thyroid nodule in Frankfurt, Germany.
The 24-year-old is not expected to return back to full match fitness until the end of the month.
Can has one assist in 10 appearances for Juve in all competitions this season.
Juventus will also be without Portuguese fullback Joao Cancelo who strained his left thigh flexors.
Additionally, Douglas Costa, Blaise Matuidi and Federico Bernardeschi are all sidelined for the match.
Chiellini and Mandzukic could however, make a return after joining team training this week.
United meanwhile could be without striker Romelu Lukaku who was absent from training at Carrington on Tuesday.
Right-back Diogo Dalot did not take part in training either for the Red Devils.
Marouane Fellaini and Antonio Valencia, who have both struggled with injuries this term did take part in team training.
A win for Juventus would guarantee qualification from Group H as winners having won all three games in the Champions League so far.
